PharmaMar(MSE:PHM)は1日、再発/難治性の多発性骨髄腫患者でデキサメタゾンとAplidin(R)(一般名プリチデプシン)の併用治療とデキサメタゾン単独治療を比較した第III相臨床試験「ADMYRE」の肯定的な主要結果を発表した。Aplidin(R)は比較対象に比べ進行、死亡のリスクの35%減少という統計的に意味のある結果を示した(p=0.0054)。この研究は主要エンドポイントを達成した。

「ADMYRE」と呼ばれるこのピボタル、無作為、オープンラベル、国際的、マルチセンター第III相臨床試験は19カ国(米国、欧州、アジア・太平洋を含む)の83医療センターで応募した255人の3回以上、6回以下の治療を受けた再発、再発・難治性の多発性骨髄腫患者を対象に行われた。

デキサメタゾンとプリチデプシンの併用とデキサメタゾン単独の効果は、IMWG(国際骨髄腫作業グループ)基準とその他の副次的効果エンドポイントを使って計算したPFS(無増悪生存期間)によって評価された。最終的なADMYREデータの全文は次の医学会議での報告のために提出される。

▽Aplidin(R)(一般名プリチデプシン)について

プリチデプシンは海洋起源の治験抗がん剤で、そのもとはアスシディアン・アプリディウム・アルビカンス(ascidian Aplidium albicans)から得られる。これは腫瘍細胞中のEF1A2を特別に標的とするクラスで初の医薬品である。プリチデプシンは現在、血液性がんの治療用に臨床開発段階であり、その中にはこの再発、難治性の多発性骨髄腫の第III相研究、再発、難治性の多発性骨髄腫でのプリチデプシン、ボルテゾミブ、デキサメタゾンの3薬併用の第I相b試験、再発、難治性の血管免疫牙性T細胞リンパ腫での第II相研究が含まれている。プリチデプシンはEMA、FDA(米食品医薬品局)からオーファン医薬品指定を受けている。

Aplidin(R) Shows Positive Results in Pivotal Phase III Clinical Trial for Multiple Myeloma

- PharmaMar intends to submit a Marketing Authorization Application to the

European Medicines Agency during the last quarter of this year

PharmaMar (MSE:PHM) today announced positive top-line results of its Phase III

clinical trial -ADMYRE- with Aplidin(R) (plitidepsin) in combination with

dexamethasone versus dexamethasone alone in patients with relapsed/refractory

multiple myeloma (MM). Aplidin(R) has shown a statistically significant 35%

reduction in the risk of progression or death over the comparator (p=0.0054).

The study has met its primary endpoint.

This pivotal, randomized, open-label, international, multicenter Phase III

clinical trial, called ADMYRE, enrolled 255 patients in 83 medical centers

across 19 countries (including the U.S, Europe and Asia-Pacific) with relapsed

or relapsed and refractory multiple myeloma after at least three but no more

than six prior therapeutic regimens.

The efficacy of plitidepsin in combination with dexamethasone versus

dexamethasone alone has been evaluated by means of PFS calculated using the

IMWG (International Myeloma Working Group) criteria and other secondary

efficacy endpoints. A full description of the final ADMYRE data will be

submitted for presentation at an upcoming medical meeting.

with TTY Biopharm in Taiwan; and with a co-promotion agreement in 8 European

countries with Chugai Pharma Europe.   

About APLIDIN(R) (plitidepsin)

Plitidepsin is an investigational anticancer agent of marine origin, originally

obtained from the ascidian Aplidium albicans. It is a first-in-class drug

specifically targeting eEF1A2 in tumor cells. Plitidepsin is currently in

clinical development for hematological cancers, including this Phase III study

in relapsed or refractory multiple myeloma, a Phase Ib trial in relapsed or

refractory multiple myeloma as a triple combination of plitidepsin, bortezomib

and dexamethasone, and a Phase II study in relapsed or refractory

angioimmunoblastic T-cell lymphoma. Plitidepsin has received orphan drug

designation by the EMA and the FDA.
