【マドリード2015年2月4日PRN＝共同通信JBN】PharmaMarは4日、米食品医薬品局（FDA）が進行性軟組織肉腫（STS）にかかった患者の治療用に同社の抗腫瘍薬YONDELIS（R、トラベクテジン）の新薬承認申請（NDA）に優先審査認定を与えたと発表した。当該STSには、アントラサイクリン系抗生物質などすでに化学療法を受けていた脂肪肉腫と平滑筋肉腫派生型を含む。これは米国でYONDELIS（R）を開発しているPharmaMarの戦略的パートナーであるヤンセン・リサーチ&ディベロップメント（Janssen Research & Development）が、2014年11月24日にNDAを申請していた（注1）。

YONDELIS（R）は2001年に欧州委員会から、2004年にFDAからそれぞれ希少疾病もしくはその症状を治療する医薬品に指定される希少疾病用医薬品指定（Orphan Drug designation）を受けた。FDAは既存の医療選択肢と比較して治療に大きな進展を示しうる医薬候補に優先審査に資格を与えている。優先審査指定は、FDAの目指すものが標準的な審査にかかる10カ月と比較して6カ月内に審査を完了するもので、NDAの評価、認定後に新薬としての処理を開始することを意味する。
YONDELIS（R）は現在、単剤（single-agent）として進行性軟組織肉腫の治療薬として77カ国で、抗悪性腫瘍製剤のペグ化リポソーマルドキソルビシンと組み合わせプラチナ感受性再発卵巣がん治療用に70カ国で製品化されている。
▽軟組織肉腫について
軟組織肉腫は筋肉、脂質、血管、神経、腱、関節部軟骨（注2）などの軟組織に発症するがんの種類である。米国ではほぼ1万2000人がSTSと診断され、2015年に約4870人がこのがんで死亡すると推定されている（注3）。平滑筋肉腫は進行性高い軟組織肉腫の1種であり、子宮、消化管、血管内皮などの平滑筋に発症する。脂肪肉腫は脂肪細胞に発症し、人体のどの部位にもある脂肪細胞で起こりうるものではあるが、最も普通に発症するのは大腿骨空洞と腹腔である（注2）。

U.S. FDA Grants Priority Review to YONDELIS(R) NDA for Advanced Soft-tissue Sarcoma

PharmaMar announces that the U.S. Food and Drug Administration (FDA) has
granted priority review to YONDELIS(R) (trabectedin) New Drug Application (NDA)
for the treatment of patients with advanced soft tissue sarcoma (STS),
including liposarcoma and leiomyosarcoma subtypes, who have received prior
chemotherapy including an anthracycline. Janssen Research & Development,
PharmaMar´s strategic partner for the development of YONDELIS(R) in the U.S.,
submitted the NDA on November 24, 2014[1].

YONDELIS(R) obtained Orphan Drug designation for STS by the European
Commission in 2001 and by the U.S. FDA in 2004, which is assigned to a drug for
treating a rare disease or condition. The FDA grants priority review status to
drug candidates that may offer major advances in treatment compared to existing
options. A priority review designation means FDA's goal is to take action,
following the validation and acceptance of the NDA, within six months as
compared to 10 months under standard review.
YONDELIS(R) is now commercialized in 77 countries for the treatment of
advanced soft-tissue sarcoma as single-agent, and in 70 countries for relapsed
platinum-sensitive ovarian cancer in combination with pegylated liposomal
doxorubicin.
About Soft Tissue Sarcoma
Soft tissue sarcomas are a type of cancer originating in the soft tissues
such as muscle, fat, blood vessels, nerves, tendons and the lining of
joints[2]. In the U.S., nearly 12,000 people will be diagnosed with STS and
approximately 4,870 are expected to die of this cancer in 2015[3].
Leiomyosarcoma is an aggressive type of soft tissue sarcoma that occurs in
smooth muscles, such as those in the uterus, gastrointestinal tract or lining
of blood vessels. Liposarcoma originates in fat cells and most commonly occurs
in the thigh and abdominal cavity, though it can occur in fat cells in any part
of the body[2].
